Genetics of Interferon Pathway Dysregulation
Type I interferons (IFN-alpha and IFN-beta) are critical antiviral cytokines whose dysregulation — either excessive or deficient — is associated with severe COVID-19, Long COVID, and autoimmune conditions. Genetic variants in interferon pathway genes may determine the IFN response amplitude.

What is Interferon Pathway Dysregulation?
The type I interferon system is the frontline antiviral defence. Upon viral RNA detection by RIG-I, MDA5 (IFIH1), and TLR7/9, IRF3 and IRF7 drive IFN-alpha/beta production, which activates hundreds of interferon-stimulated genes (ISGs) through STAT1/2 signalling. Deficient IFN responses (e.g. rare TLR7 loss-of-function variants in young men) predispose to severe COVID-19. Conversely, excessive or persistent IFN signalling — as seen in Aicardi-Goutières syndrome and lupus — causes systemic inflammation. Long COVID patients show evidence of persistent IFN dysregulation and elevated IFN signatures in some studies.
Biological pathways commonly discussed in relation to Interferon Pathway Dysregulation
Research into Type I Interferon Signalling and Interferonopathy has highlighted several biological pathways that may be relevant. Genetic variants within these pathways may influence individual susceptibility, symptom severity, or the trajectory of the condition.
Type I Interferon
IFN-alpha (multiple IFNA subtypes) and IFN-beta (IFNB1) bind IFNAR1/2 to signal through JAK1/TYK2 and STAT1/2. The strength and duration of IFN signalling depends on feedback inhibition (SOCS1/3, USP18) and genetic regulation of receptor sensitivity.
Rig I Mda5
RIG-I (DDX58) and MDA5 (IFIH1) are cytosolic RNA sensors detecting viral dsRNA. IFIH1 variants are associated with type 1 diabetes, Aicardi-Goutières syndrome, and altered COVID-19 susceptibility. DDX58 variants have been linked to IFN pathway hyperactivation.
Jak Stat Signalling
JAK1 and TYK2 phosphorylate STAT1 and STAT2 downstream of interferon receptors. TYK2 P1104A is a common coding variant protective against autoimmune diseases, reducing TYK2 catalytic activity.
Isg Expression
Interferon-stimulated genes including IFIT1, OAS1, MX1, and IFITM3 confer direct antiviral protection. IFITM3 rs12252 affects restriction of influenza, SARS-CoV-2, and other respiratory viruses.

How personal DNA data may provide educational insights
Consumer DNA files from 23andMe, AncestryDNA, and MyHeritage contain raw SNP data. When you upload your file to ChronicDNA, the analysis engine checks your genotype against a curated panel of 80+ genes selected for their relevance to Long COVID, ME/CFS, and connected biological pathways - some of which may relate to areas such as Interferon Pathway Dysregulation.
This is not a diagnostic service. Genetic variants do not determine whether you have Interferon Pathway Dysregulation, nor do they predict your clinical outcomes. The report is educational - it explains which variants you carry, what the research says about them, and how they fit into the biological pathways most relevant to chronic illness. All results should be discussed with a qualified healthcare provider.

Frequently asked questions
How do interferon pathway variants relate to severe COVID-19?
Rare loss-of-function variants in TLR7 have been found at higher frequency in young men with severe COVID-19, implicating deficient IFN-alpha production. Common variants in IRF7 and IFNAR2 were identified in GWAS of severe COVID-19 hospitalization. The IFN response amplitude appears critical in determining COVID-19 severity.
What is an interferonopathy?
Interferonopathies are genetic conditions characterized by constitutive upregulation of the type I interferon pathway, producing chronic systemic inflammation resembling lupus. Examples include Aicardi-Goutières syndrome (TREX1, RNASEH2 variants) and STING-associated vasculopathy. Milder variants in interferon pathway genes may contribute to autoimmune susceptibility.
Does Long COVID involve interferon dysregulation?
Studies have documented elevated type I IFN signatures in some Long COVID patients months after acute infection. This persistent IFN activation may reflect ongoing innate immune stimulation and has been proposed as one mechanism underlying fatigue and cognitive symptoms.
Which interferon pathway genes are studied?
Key genes include TLR7, IFIH1 (MDA5), IRF3, IRF7, STAT1, STAT2, TYK2, OAS1, MX1, IFITM3, and IFNAR1/2. Together these cover viral sensing, IFN production, JAK-STAT signalling, and ISG effector function.
