Genetics of Oxidative Stress
Oxidative stress occurs when the production of reactive oxygen species (ROS) exceeds the body's antioxidant capacity, damaging cells, proteins, and DNA. Genetic variants in antioxidant defence genes may influence individual susceptibility to oxidative damage.

What is Oxidative Stress?
Every cell produces reactive oxygen species as a byproduct of normal metabolism, particularly in the mitochondria. Under healthy conditions, antioxidant enzymes such as superoxide dismutase (SOD2), glutathione peroxidase (GPX), and catalase neutralise these reactive molecules. When this balance is disrupted — either through excessive ROS production or impaired antioxidant capacity — oxidative stress results. Chronic oxidative stress drives cellular damage, inflammation, mitochondrial dysfunction, and accelerated ageing. It has been identified as a contributing factor in Long COVID, ME/CFS, cardiovascular disease, and neurological conditions.
Biological pathways commonly discussed in relation to Oxidative Stress
Research into Oxidative Stress and Antioxidant Defence has highlighted several biological pathways that may be relevant. Genetic variants within these pathways may influence individual susceptibility, symptom severity, or the trajectory of the condition.
Antioxidant Defence
Antioxidant enzymes including SOD2, GPX1, CAT, and PRDX family members neutralise reactive oxygen species. Genetic variants reducing enzyme activity may increase cellular vulnerability to oxidative damage.
Mitochondrial Ros
Mitochondria are the primary source of intracellular ROS. Variants affecting mitochondrial electron transport chain efficiency may increase ROS leak and oxidative burden.
Nrf2 Pathway
NRF2 (encoded by NFE2L2) is the master regulator of antioxidant gene expression. Variants affecting NRF2 activity can reduce the overall antioxidant response capacity of cells.
Glutathione Metabolism
Glutathione is the body's primary endogenous antioxidant. Variants in GST genes, GCL, and related enzymes affect glutathione synthesis and recycling, influencing cellular redox status.

How personal DNA data may provide educational insights
Consumer DNA files from 23andMe, AncestryDNA, and MyHeritage contain raw SNP data. When you upload your file to ChronicDNA, the analysis engine checks your genotype against a curated panel of 80+ genes selected for their relevance to Long COVID, ME/CFS, and connected biological pathways - some of which may relate to areas such as Oxidative Stress.
This is not a diagnostic service. Genetic variants do not determine whether you have Oxidative Stress, nor do they predict your clinical outcomes. The report is educational - it explains which variants you carry, what the research says about them, and how they fit into the biological pathways most relevant to chronic illness. All results should be discussed with a qualified healthcare provider.

Frequently asked questions
What is oxidative stress?
Oxidative stress occurs when reactive oxygen species (ROS) accumulate faster than antioxidant systems can neutralise them, damaging cellular components including DNA, proteins, and lipids. It is implicated in ageing, inflammation, and chronic illness.
How may oxidative stress genetics relate to Long COVID or ME/CFS?
Elevated oxidative stress markers have been reported in Long COVID and ME/CFS patients. Genetic variants reducing antioxidant capacity — particularly in NRF2, SOD2, or glutathione pathway genes — may influence individual vulnerability to oxidative damage during and after infection.
Which genes are studied in relation to oxidative stress?
Key genes include NFE2L2 (NRF2), SOD2, GPX1, CAT, GSTM1, GSTT1, NQO1, and HMOX1. These encode the primary antioxidant enzymes and the transcription factors that regulate them.
Can consumer DNA data show oxidative stress variants?
Yes — consumer DNA files from 23andMe, AncestryDNA, and MyHeritage include common SNPs in antioxidant genes such as GSTM1, SOD2, and NFE2L2. ChronicDNA analyses these variants for educational insights.
